The discovery and engineering of novel fluorescent proteins (FPs) from diverse organisms is yielding fluorophores with exceptional characteristics for live‐cell imaging. In particular, the development of FPs for fluorescence (or Förster) resonance energy transfer (FRET) microscopy is providing important tools for monitoring dynamic protein interactions inside living cells. The increased interest in FRET microscopy has driven the development of many different methods to measure FRET. However, the interpretation of FRET measurements is complicated by several factors including the high fluorescence background, (...) the potential for photoconversion artifacts and the relatively low dynamic range afforded by this technique. Here, we describe the advantages and disadvantages of four methods commonly used in FRET microscopy. We then discuss the selection of FPs for the different FRET methods, identifying the most useful FP candidates for FRET microscopy. The recent success in expanding the FP color palette offers the opportunity to explore new FRET pairs. (shrink)

Talents that selectively facilitate the acquisition of high levels of skill are said to be present in some children but not others. The evidence for this includes biological correlates of specific abilities, certain rare abilities in autistic savants, and the seemingly spontaneous emergence of exceptional abilities in young children, but there is also contrary evidence indicating an absence of early precursors of high skill levels. An analysis of positive and negative evidence and arguments suggests that differences in early experiences, preferences, (...) opportunities, habits, training, and practice are the real determinants of excellence. This Response addresses eight issues raised in the commentaries: (1) the question of how innate talents should be defined; (2) relationships between the talent account and broader views concerning genetic variability; (3) the quality of the empirical evidence for and against the talent account; (4) the possible involvement of innate influences on specific abilities; (5) the possibility of talent-like phenomena in autistic savants; (6) alternative explanations of exceptional expertise at skills; (7) practical and educational implications of the talent account and (...) alternative positions. A striking thesis lies at the core of Davidson's philosophy: when we attribute intentional content to another creature's mental states and speech acts, we must treat the creature as largely conforming to our own rational norms. I will discuss how this thesis informs Davidson's treatment of rationality and intentionality. After reviewing some historical background, I present basic aspects of Davidson's position. I then examine various worries about the position. I conclude by highlighting some key Davidsonian insights into (...) rationality. (shrink)

Peter Abelard (1079–1142 ce) was the most wide‐ranging philosopher of the twelfth century. He quickly established himself as a leading teacher of logic in and near Paris shortly after 1100. After his affair with Heloise, and his subsequent castration, Abelard became a monk, but he returned to teaching in the Paris schools until 1140, when his work was condemned by a Church Council at Sens. His logical writings were based around discussion of the “Old Logic”: Porphyry's Isagoge, aristotle'S Categories and (...) On Interpretation and boethius'S textbook on topical inference. They comprise a freestanding Dialectica (“Logic”; probably c.1116), a set of commentaries (known as the Logica [Ingredientibus], c. 1119) and a later (c. 1125) commentary on the Isagoge (Logica Nostrorum Petititoni Sociorum or Glossulae). In a work Abelard called his Theologia, issued in three main versions (between 1120 and c.1134), he attempted a logical analysis of trinitarian relations and explored the philosophical problems surrounding God's claims to omnipotence and omniscience. The Collationes (“Debates,” also known as “Dialogue between a Christian, a Philosopher and a Jew”; probably c.1130) present a rational investigation into the nature of the highest good, in which the Christian and the Philosopher (who seems to be modeled on a philosopher of pagan antiquity) are remarkably in agreement. The unfinished Scito teipsum (“Know thyself,” also known as the “Ethics”; c.1138) analyses moral action. (shrink)

Donald Davidson's unified theory of language and action can be understood as a continuation of the traditional analytic approach within philosophy of language and action. In the following we give an outline of some of the main themes of his philosophy centered around his core theory of radical interpretation (RI). His theory of rationality, his thesis of the anomalousness of the mental, and the externalist approach in his model of triangulation all follow in a systematic way from RI. Yet, (...) Davidson's philosophy can also be seen as giving a new systematic elaboration of central perspectives of W,v.O Quine's theory of language. The laboratory animal environment: room for concern.

The question of when or if a nation should intervene in another country’s affairs is one of the most important concerns in today’s volatile world. Taking John Stuart Mill’s famous 1859 essay “A Few Words on Non-Intervention” as his starting point, international relations scholar Michael W. Doyle addresses the thorny issue of when a state’s sovereignty should be respected and when it should be overridden or disregarded by other states in the name of humanitarian protection, national self-determination, or national (...) security. In this time of complex social and political interplay and increasingly sophisticated and deadly weaponry, Doyle reinvigorates Mill’s principles for a new era while assessing the new United Nations doctrine of responsibility to protect. In the twenty-first century, intervention can take many forms: military and economic, unilateral and multilateral. The notion of ontography is characterizable as open-source, both due to its collaborative development, its heterogeneous backgrounds and its broad applicability. In my paper, I concretize these open-source aspects of ontography firstly by redefining it with reference to E. Winkler’s dialogue Die Erkundung der Linie and secondly by applying it to the Gestalttheoretical topics of figure-ground reversals and bidirectional part-whole relations.

Key sections of Walter Benjamin's montage-text Berlin Childhood around 1900 figure the relationship between human experience and modern media, with the sections that frame the text, ‘Loggias’ and ‘The Moon’, structured around metaphors of photography. Drawing on the work of Siegfried Kracauer, and especially his seminal essay ‘Photography’, Benjamin develops, in the course of his book, a theory of photography's relationship to experience that runs counter to the better-known theories developed in such essays as ‘Little History of Photography’ and ‘The (...) Work of Art in the Age of its Technological Reproducibility’, theories that are part of the broad currents of technological utopianism and, as such, emphasize photography's transformative potentials. In the Berlin Childhood, Benjamin instead emphasizes photography's role in the mortification and annihilation of meaningful human experience. Machine learning algorithms pervade contemporary society. They are integral to social institutions, inform processes of governance, and animate the mundane technologies of daily life. Consistently, the outcomes of machine learning reflect, reproduce, and amplify structural inequalities. The field of fair machine learning has emerged in response, developing mathematical techniques that increase fairness based on anti-classification, classification parity, and calibration standards. In practice, these computational correctives invariably fall short, operating from an algorithmic idealism that does not, and cannot, address systemic, Intersectional (...) stratifications. Taking present fair machine learning methods as our point of departure, we suggest instead the notion and practice of algorithmic reparation. Rooted in theories of Intersectionality, reparative algorithms name, unmask, and undo allocative and representational harms as they materialize in sociotechnical form. We propose algorithmic reparation as a foundation for building, evaluating, adjusting, and when necessary, omitting and eradicating machine learning systems. (shrink)

Despite his seminal role in the history of philosophy, the thirteenth century thinker Albert the Great remains little known. Prior to World War II, his massive literary output was not fully analyzed by historians largely because, as Etienne Gilson put it, of the amazing "amount of philosophical and scientific information heaped up in his writings." After the war, Albert's work began to receive more attention. By 1955, the Louvain medievalist Fernand Van Steenberghen could confidently declare that Albert was the first (...) thinker to establish "the rightful place of learning in Christianity." A decade later, James A. Weisheipl uncovered evidence of Albert's distinctively naturalistic interpretation of Aristotle in .. (shrink)

Proposals for a universal basic income have reemerged in public discourse for a variety of reasons. Marx’s critique of exploitation suggests two apparently opposed positions on a basic income. On the one hand, a basic income funded from taxes on labor would appear to be exploitative of workers. On the other hand, a basic income liberates everyone from the vulnerable condition in which one is forced to sell one’s labor in order to survive, and so seems to be one way (...) of abolishing exploitation at its root. This paper will develop a conception of exploitation that resolves the conflict in favor of basic income. The conception of exploitation is grounded in a liberal egalitarian conception of justice rather than in Marx’s labor theory of value or an exclusive focus on the worker-capitalist relation. This position is not premised on an acceptance of the basic institutions of capitalism, but rather is a standpoint from which to evaluate them. It is not necessary to downsize our ideas of freedom and equality. But it is less obvious than it appeared in classic Marxist formulations that socialism is necessary for social justice. To quote the title of a famous article, there could be a ‘capitalist road to communism’, if a substantial basic income is feasible in a capitalist society. (shrink)

The metaphysical language of impermeable surfaces and levels, and rigid hierarchies, is consonant in James's writing with the assumption of what Dewey calls an individual/society split. ;Chapter Two focuses upon the relational self from the Principles of Psychology. The central pair of terms is that of strength/fragility, in which a self is revealed that is both functionally efficacious through activities of emphasis, selection, and negation, and permeable (...) to context. ;Chapter Three discusses the "social organism" and the "intellectual republic" from The Will to Believe and Other Essays in Popular Philosophy. James uses more socially conscious metaphors, and discusses the public and the private; "inner" and "outer" tolerance; and the "personal contribution," "X," which must be added to the "world," "M." ;Chapter Four discusses the "nucleus" of relations from Essays in Radical Empiricism. The personal aspect of social experience falls within the realm of "co-conscious relations" and the public falls within the sphere of the "conterminous." The strong side of the self, from Chapter Two; the private, or personal; inner tolerance; and the personal contribution, "X," from Chapter Three, belong to the sphere of co-conscious relations, as discussed in Chapter Four, just as the fragile side, the public; outer tolerance; and the world, "M," all belong to the sphere of conterminous relations. ;Chapter Five discusses the metaphor of Papini's corridor from Pragmatism. The adjoining rooms can be seen to symbolize the sphere of co-conscious relations, and the corridor of conterminous ones. The metaphysics of background and foreground points to an instrumentalist interpretation of the self, which represents a complete departure from James's early individualism. (shrink)
